To insure future RFET firmware upgrades, we need to provide an option to access COM1 (UART1) on our custom board. Please provide specs for this asap. Electrical drawings would be great too, as an example.

You should be able to get what you need from the existing Hardware Reference Manual, which comes with all EK2500 Evaluation kits (look on the CD).

I think the RF Engine datasheet is also available on the CD, but you could also download it from http://www.synapse-wireless.com/documents/products/RFEdsW.pdf
To summarize the above resources, you are interested in RF Engine pins 9 and 10, and possibly 11 and 12.
RFE pin 9 is GPIO7 AKA RX_UART1 (3.3 volt logic level INPUT)
RFE pin 10 is GPIO8 AKA TX_UART1 (3.3 volt logic level OUTPUT)
RFE pin 11 is GPIO9 AKA CTS1 (3.3 volt logic level input)
RFE pin 12 is GPIO10 AKA RTS1 (3.3 volt logic level output)
If you want the first UART (UART0) then the pin numbers of interest are 5-8 instead of 9-12.

Is that necessary to use pins 11, 12 (CTS, RTS) for firmware update?

The RTS and CTS pins are not currently used by the firmware updater
